All lanes between Junction Six for Godstone, Surrey and Junction Five for Sevenoaks, Kent are closed.
Traffic cameras show long stretches of the empty motorway.
On Twitter, National Highways South East (NHSE) has written: "Traffic on the #M25 between J6 #Godstone and J5 #M26 #Sevenoaks is currently stopped in both directions due to a police led incident. Delays are beginning to build on approach."
In a second post, NHSE notes: "The #M25 in #Kent has now been closed anti-clockwise between J6 (#Godstone) and J5 (#M26 / #Sevenoaks) due to a Police incident. The clockwise carriageway remains blocked."
Kent Police was called at 7.40pm after a pedestrian fell and suffered serious injuries.
A spokesman for the force said no vehicles were involved.
He added: "The anti-clockwise carriageway is currently closed between junctions 6 and 5.
"There are also delays on the clockwise carriageway. Officers and ambulance crews remain at the scene."
Highways England expects the route to clear between 10.45pm and 11pm tonight with traffic returning to normal over the same period.
It is warning of delays upwards of 30 minutes.
In an update at 9.10pm, NHSE announced that the clockwise carriageway reopened althought anti-clockwise the route remains closed as the emergency services work at the scene.
