Pictures showed police, ambulances and fire crews at the scene on the seafront.
Sleepy guests were reportedly told to stand outside after a fire alarm started ringing out at 6.30am.
It is understood an electrical fault could be causing the problem but this hasn't been confirmed yet.
Labour have been holding their five-day conference in Brighton this week.
Sir Keir Starmer is due to give a keynote speech - his first in-person conference address - at midday to mark the end of the event.
Queen's Hotel has hosted six fringe events for the conference - including speeches from Kim Leadbeater, Liz Kendall and Ian Murray.
The Sun Online has contacted Sussex Police for comment.
